The differentiation of brewing yeasts and the detection of wild yeasts based on cultivation often causes difficulties because of high similarities amongst brewing yeasts and between brewing and wild yeasts respectively. To get a more rapid and specific detection of these organisms different Real-Time PCR methods were developed. Using Real-Time PCR it was possible to establish assays for the differentiation of bottom-and top-fermenting yeasts. Moreover specific assays were designed for the detection of harmful wild yeasts, e.g. S. Diastaticus, Dekkera spp. Combining PCR with selective cultivation (MYPG+CuSO CuSO4) preexisting know-how was successfully combined with PCR and identification accuracy was improved.
