Mastitis is the most common infectious disease affecting dairy cows and is the most economically important disease of the dairy industry. Identification of bacterial pathogens in milk from a cow with mastitis is regarded as the definitive diagnosis ofintramammary infections. Current methods for routine identification and confirmation of mastitis pathogens requires a minimum of 48 hours. Identification of bacteria utilizing DNA-based technology could eliminate analysis of phenotypic characteristics utilizing biochemical tests, serotyping and enzymatic profiles and provide an identification in 24 hours or less. Real-Time Polymerase Chain Reaction (PCR) is a relatively new DNA based technique that monitors amplification of target DNA in real-time by monitoring florescence. The objective of this study was to develop a Real-Time PCR procedure for detection of Staphylococcus aureus, Streptococcus agalactiae, Streptococcus uberis, Streptococcus dysgalactiae subsp dysgalactiae and Listeria monocytogenes directly from milk.
