Ortho Ester-Based Acid-Labile Cationic Lipids for Gene Delivery

Several ortho ester-based pH-sensitive cationic lipids were developed. Their lipoplexes mediated more efficient gene transfection than their pH-insensitive controls in both cultured cells and in mice. The physicochemical characterizations of the lipoplexes support the hypothesis that the pH-triggered endosome destabilization is responsible for the enhanced transfection.
