A wheat microsatellite consensus map for breeding and genomics

Large-scale molecular breeding in crops requires two fundamental genetic resources: a high-density molecular genetic map and access to molecular markers that are amenable to high throughput technology. There have been several public and private efforts to develop microsatellite markers in wheat and today, large collections of microsatellite markers are being released to the public. This report brings together most of the major microsatellite markers available in the public domain to develop a single,consensus map of wheat with > 1,000 microsatellite loci mapped. All of the mapping was done in four populations: Synthetic x Opata (RIL), RL4452 x AC Domain (DH), Superb x BW278 (DH) and Wuhan x Maringa (DH). The microsatellite marker collection includes WMC (Agrogene, Isaac), GWM (IPK, Roder), GDM (IPK, Roder), BARC (USDA, Cregan), CFD (INRA, Sourdille) and CFA (INRA, Sourdille). Map maker V3.0 was used to construct individual genetic maps at LOD>2.5 with a maximum recombination of 0.3. The Syntheticx Opata population was the most densely mapped and was used predominantly in determining marker order. JoinMap V3.0 was used to create the consensus map at LOD>2.5. The final map will be displayed, access to primer sequences, allele sizes and use of themap in molecular breeding will be described.
