Warnings from diagnostic laboratories and test kit manufacturers dictate the need for perfect blood sample handling: immediate chilling, no hemolysis, prompt serum-clot separation, and ultra-low temperature (-80°C) serum storage (Lippi et al., 2006)in order to produce reliable and accurate results. However, between the time of collection and subsequent disease antibody quantification, perfect blood sample handling rarely occurs given the daily vagaries of swine medicine practice. Even though sample handling has conventionally been thought to affect the outcome of diagnostic tests, studies assessing the effect of poor sample handling on modern, ELISA-based serum antibody assays are lacking.
