Potent cyclic angiotensin II analogues confirm the ring cluster conformation:implications in the design of AT1 non-peptide antagonists

Accumulated experimental evidence for Angiotensin II supports bioactive conformation characterized by a charge relay system between Tyr hydroxyl,His imidazole and Phe carboxylate,analogous to that found in serine proteases,as well as a ring cluster of the triad key aminoacids Tyr~4-His~6-Phe~8 which appears to be responsible for activity [1,2].
