An exogenous gene (green fluorescent protein: GFP) directly introduced into thetestis of adult male chicken was successfully transferred to the eggs (F0) fertilized by spermatozoa from treated males. After F0 chickens were raised to sexual maturation, F0 male chickens with normal female, and F0 male chickens with F0 female chickens were subjected to artificial insemination according to routine method. The collected fertile eggs were incubated for 72h to obtain F1 generation and the F1 embryos were examined for the detection of introduced DNA. The expression of GFP gene in the F1 embryos was observed under a fluorescent microscope. Furthermore, the presence of GFP gene in the embryo extracts was confirmed by PCR analysis. These results suggest thatexogenously introduced DNA was successfully transferred to the F1 generation of the chicken.
