PROCEDURE FOR THE DIRECT TRANSFER AND STABLE INTEGRATION OF FOREIGN DNA IN HEREDITARY PROTOPLAST MATERIAL IN PLANTS, AND FOR THE EXPRESSION AND REPLICATION OF THIS FOREIGN DNA.
Transformation of the hereditary material of plants comprises transferring a gene (under control of plant expression signals) directly into plant cells without the aid of a natural infective system. Pref. foreign genes and plant protoplasts are mixed in a soln. (esp. an osmotically-balanced protoplast culture medium) and allowed to stand until the genes have been taken up. The gene can be of plant, animal, microbial, viral or synthetic origin, and the expression signals of plant, animal or synthetic origin. The gene can be combined with replication or integration signals, and opt. with an enhancer sequence (most pref. all 3). It can also include a selection marker. The plants being transformed are esp. of the families Solanaceae, Cruciferae and Gramineae.
