It is reported that when mouse cells are exposed to a pressure of 80 MPa, the cells in S-phase are sensitive to the pressure. To analyze the sensitivity of S-phase cells to pressure, we used a Xenopus egg cell-free system. The DNA replication was monitored by the incorporation of biotin-16-dUTP into sperm nuclei. The progress of DNA replication was completely inhibited by exposure to a pressure (80MPa) of a mixture of sperm nuclei and egg extracts or egg extracts only. On the other hand, 80 MPa-treated sperm nuclei incorporated biotin-16-dUTP, and entered M-phase. These results suggest that sperm nuclei are insensitive to a pressure of 80 MPa and some components in egg extracts are sensitive to the pressure.
