It has been demonstrated in several plant species that the lumen of the chloroplast thylakoids may be filled with an electron dense (densely staining) substance (DS), while the thylakoid membrane is lightly staining (Fig. 1, for references see Keresztes and Sarvari). We carried out fixation and extraction probes and studied the effect of experimental conditions for further elucidating these phenomena. Materials used were Helianthus annuus cotyledons and Perilla frutescens leaves.
