Transforming indica rice cuttivars grown in Vietnam using Agrobacterium tumefaciens or particle bombardment

Genetic transformation mediated by Agrobacterium tumefaciens was performed in four indica cuitivars grown in Vietnam and in two japonica cuitivars. Embryogenic calli derived from mature seeds were inoculated with A. tumefaciens strain LBA4404, in which the T-DNA contained CaMV 35S-iox-hyg-lox-gusA or Ubi-Cre-CaMV 35S-hyg or Ubi-Cre-CaMV 35S-barB. Transformation efficiency, which is recorded as the number of independent transgenic plants showing Southern blot~+ per number of caili inoculated, was 2.2-10.1% for indica cuitivars and 10.4-25.8% for japonica cuitivars, Southern analyses of T_0 transgenic. plants and T_1 and T_2 progenies revealed that most of the transgenes were integrated in the genome as a single locus and segregated following Mendeiian inheritance. By using particle bombardment, we cotransferred the two insecticidal genes crylAc and GNA in scented indica variety KDM105. The transgenic T_0 plants expressing both genes were identified by western blot analysis. In the T_4 generation, afew lines showed resistance to brown planthopper in the seed-box test.
