Subcutaneous implantation of a bone morphogenetic protein (BMP) - collagen type I composites induced direct bone formation. We then conducted an ultrastructural analysis of this bone formation. The composite induced the bone without cartilage formation within 14 days after implantation. An initial calcification was observed not only on matrix vesicles within osteoid the tissue, but also on BMP- containing proteins and carrier collagen fibrils. These results demonstrate that the initial calcification of direct bone formation induced by BMP appears at multiple-nucleation sites, and carriers not only stabilizes BMP, but also play a role as the site of the bone in nucleation and calcification.
