Transcriptional Regulation Analysis of a Novel Human Lung Adenocarcinoma Metastasis-related Gene (MAR VELD1)

Lung carcinoma,as one of the leading cause of cancer deaths worldwide,has not been fully understood yet in its mechanism.Here we report the transcriptional regulation of a potential human lung adenocarcinoma gene——MARVELDI.Bioinformatic analysis of its promoter region indicated that there are no TATA or CAAT box,but some potential GC boxes.Further studies by the Dual-Luciferase Reporter System and EMSA,showed that the first GC box (-73bp~78bp) of its promoter region has relatively higher transcriptional activity,and could be regulated by transcription factor Sp1 and Sp3.Sp1,as an positive factor,could increase the transcription activity.In contrast to Sp1,Sp3 showed suppressive activity on transcription induced by Sp1.The region between -243bp and -257bp,also showed remarkably repressive activity on the transcription,although no obvious cis element was found at this region at present.
