DNA enzymes are a novel therapeutic tool for disease intervention at the molecular level. Two DNA enzymes were designed to target the template region of the human telomerase RNA (hTR) component. Activity of the DNA enzymes was demonstrated by in vitro cleavage reactions. Subsequently, they were shown to inhibit telomerase activity in cells. To facilitate cellular delivery, two delivery strategies, cationic lipids and PLGA microspheres, were examimned.
