The efficacy of many protein and DNA therapeutics is limited by nonproductive intracellular trafficking. We have developed a polymer-based approach to manipulating the intracellular trafficking of therapeutics that is inspired by biological polymers, i.e. proteins, that circumvent vesicular trafficking pathways by enhancing transport across the endosomal membrane. These pH-responsive polymers enhance the delivery of DNA and proteins into the cytoplasm, and initial animal studies show that they can enhance gene delivery in a local delivery wound healing model..
