In order to overcome the disadvantages caused by the flow principle used in automatic counting and classification of blood cells, a new method of counting and classification of blood cells is presented in this paper. This method is a laser light scattering method from stationary suspensions. The blood cell is simulated by an isotropic sphere, and Mie scattering theory is used to describe the scattering of blood cells. In order to get the size distribution of blood cells from their scattered light intensity distribution, the NNLS method combined with Powell method and precision punishment method are used. The dependence between blood cell concentration and its scattered light intensity is obtained from experiment. It is shown that there exists a linear relationship between them. By this new method, the size distribution of red blood cells and white blood cells are obtained, and the white blood cells can be classified as lymphocyte, middle-sized cells and neutrocyte.
