Microencapsulated Hepatocytes Hemoperfusion for the Treatment of Fulminant Hepatic Failure Rats

Hepatocytes were isolated from suckling porcine using modified seglen's collagenase perfusion technique, and were encapsulated with alginate-polylysine-alginate (APA) microcapsules. Fulminant hepatic failure(FHF) was induced in male SD rats by galactosamine. Hemoperfusion performed 24hrs after galactosamine injection. In vitro culture, both encapsulated hepatocytes and free hepatocytes express the ability to synthesis Albumin, Urea for 7 days, Encapsulated hepatocytesperfusied with Williams' E medium was shown to provide albumin, urea synthesis functions in 24hrs. The survival time after treatment of microencapsulated hepatocytes hemoperfusion group was longer than other groups, and Serum ALT, TBIL levels decreased.
