Nephroprotective Effect of Ethanolic Extract Vernonia Amygdalina Delile on Rats

Doxorubicin causes nephrotoxicity by decreasing the glomerular filtration rate. Various mechanisms contribute to renal dysfunction after induced doxorubicin. This study aims to determine the nephroprotective effect of ethanol extracts of African leaves (EEAL) to reduce creatinine, urea, and uric acid levels in doxorubicin-induced rats. In vivo test using 24 rats divided into 6 groups which were normal, negative control (Doxorubicin 20 mg / kgBW), positive control (Doxorubicin 20 mg / kgBW + Vitamin E 1%), Group I (Doxorubisin 20 mg / kgBW + 100 mg / kg EEAL), Group II (Doxorubicin 20 mg / kgBB + 300 mg / kgBW), and Group III (Doxorubicin 20 mg / kgBB + 500 mg / kgBB). Doxorubicin at a dose of 5 mg / kg body weight given on days 1, 7, 14, 21. The EEAL was given for 20 days, on day 21 the rats were anesthetized using ketamine at a dose of 70 mg / kg body weight i.v. The rats blood was taken from the heart, then centrifuged for 10 minutes at a speed of 3000-4000 rpm. Then, urea, creatinine, and uric acid were measured. The results showed that EEAL decreased urea, creatinine, and uric levels in rats that were only induced by doxorubicin such that the EEAL has nephroprotective effect.
