Isolation of antigen-specific plasma cells using Janus particles

Larger silica particles demonstrated to be the best combination to collect low concentration of antibody in cell culture and cell supernatant. Biological silica Janus particles were successfully conjugated with different anti-CD molecules and Protein G. Targeting and collection of secreted antibody was demonstrated using Janus and heterofunctional particles. Comparison of different sizes and materials of Janus particles demonstrated that larger silica Janus particles is the best combination to obtain higher efficiency of targeted cells and collection of secreted antibody. Janus particles demonstrated higher cells targeted, while heterofunctional particles demonstrated higher antibody collection and better localization. Future studies will improve collection of secreted antibodies by minimizing the detection of antigen on unbound particles. Finally, ASCs will be purified from human blood using the best conditions.
