The main purpose of this paper is to study the effects of extracellular matrix (ECM) protein micropatterns with different pattern shape and size on polystyrene surfaces fabricated by microcontact printing on human osteoblasts adhesion, spread and protein expression. In order to investigate the adhesion, spread and protein expression of human osteoblasts, we stain the nucleolus and cytoskeleton with DAPI and phalloidine respectively, and the type Ⅰ collagen and osteocalcin were labeled by immunohistochemical method. The laser confocal scanning microscopy was used to examine all of these results. The results indicated that the micropattemed bioactive surfaces have significant influence on cell adhesion, spread, alignment and functions. The cells preferentially adhered on micropattern protein zones. The micropattern shape and its sizes affected not only the cell adhesion morphology and spread degree but also the type Ⅰ collagen and osteocalcin expression. The spread behavior of cell has some positive connections with protein expression and the cell with more spread expressed more type Ⅰ collagen and osteocalcin. The results of this paper suggested that the ECM protein micropatterns fabricated in the polystyrene can effectively regulate human osteoblasts growth and functions.
