The high fuel ethanol cost was predominantly attributed to larger enzyme dosage, lower enzyme activity and lower hydrolysis yield. In this article, comparison of cellulolytic enzyme profiles between Trichoderma reesei cellulase with lower enzyme activity (2.68 FPIU/ml) and commercial cellulase with higher enzyme activity (143.20 FPIU/ml) was investigated. The retained and permeated fractions from ultrafiltration of T. reesei cellulase and commercial cellulase were submitted to ion-exchange chromatography respectively, and each fraction was subjected to SDS-PAGE. β - glucosidase, cellobiohydrolase, endoglucanase and xylanase between T. reesei cellulase and commercial cellulase exhibited differences in terms of types, distribution and amount respectively. The spectra of T. reesei cellulase and commercial cellulase on 2-DE displayed that in the 20kDa region (pIapp. 5) (identified as XYNⅠ), 25-35kDa region (pIapp. 7) (contained Cel 12A) and 45-70kDa region (pIapp. 7), there were some enzymatic protein spots in T. reesei cellulase but those were absent in commercial cellulase.
