To provide data for studies on avian disease resistance, Tianfu goose interferon-alpha (tf-GoIFNa) was amplified from con A stimulated peripheral blood mononuclear cells by RT-PCR. The open reading frame (ORF) of the tf-GoIFN-a containing 576bp nucleotides was preliminarily identified by the software of BlastN and ORF Finder. Specific primers were designed and PCR products containing this ORF were cloned into the vector of PMD18-T. The molecular characteristic of the tf-GoIFN-a gene was then analyzed with bioinformatics software. The tf-GoIFN-a showed, respectively, approximately 98%, 96%, 68% similarities in cDNA, and also showed, respectively, approximately 95%, 92%, 48% similarities in amino acid sequence to Dongbei goose, duck and chicken IFNa. A phylogenetic tree of the interferon gene family defined the evolutionary relationship of various species. Phylogenetic tree analysis showed that fifthteen interferons clustered together in a species-specific manner and fell into three large groups, in addition to the tf-GoIFN-a was closely related to Dongbei goose, but to grass carp, they were divided into two different clades, with high bootstrap support
