An efficient protocol for direct regenerationof blackberry (Rubus hybrid) cv.‘K iowa' (PI No.638183) from immature leaflet explants excised from axenic seedlings has been developed.Initially,the leaflets from 5-week-old mother cultures on MS medium were used as the explants.Treatments on MS medium with 21 days' dark treatment and from the 1st (from top) leaflets revealed better regeneration effects.Cultures on medium MS+ forchlorfenuron (CPPU) 1.0 mg·l-1+indole-3-acetic acid (IAA) 0.5 mg·l-1 produced the highest regeneration frequency (95.00%); The best proliferation medium (coefficient 4.70) was MS+ N6-benzylaminopurine (BAP) 0.5 mg·l-1 +IAA 0.1 rag·1-1.The optimum rooting medium was 1/2 MS+a-naphthalene acetic acid (NAA) 0.03 mg· 1-1.The results were valuable for rapid propagation and genetic transformation of blackberry.
