In order to obtain the best culture modes and growth hormone on rooting of walnut, several measures to promote shoots rooting of walnut in vitro were performed in this study. It was found that successful rooting was closely related to the method of cultivation, concentration level and treatment time of growth hormone, and culture condition in dark. Our data indicated that using two-step method, which is firstly induced on 1/4DKW with IBA (2.0-10.0) mg/L for 6~15 days, then transferred into the 1/4 DKW without endogenous hormone and growth regulator and incubated 12~15days under a dark condition, the rooting rate of in vitro plantlets will increase obviously.
