Nanoparticles-in-microsphere oral system (NiMOS), previously optimized for particle size using a 3~3 full factorial design, were used for encapsulation and oral delivery of plasmid DNA encoding for enhanced green fluorescent protein (pEGFP-N1). Upon administration in Balb/c mice, NiMOS were able to successfully transfect in the small and large intestine and the protein expression was observed for up to 5 days following a single 100 μg dose.
