The aim of the present research work was to develop and optimize a vaginal drug delivery system of a Novel Antimicrobial Combination (NAC) from herbal sources that would yield a prolonged local antimicrobial effect in a sustained release manner. Xanthan gum gels of the NAC were prepared in natural honey base and optimized using Box-Behnken statistical screening design. Concentration of xanthan gum, carboxymethyl cellulose (CMC), and honey were selected as independent variables. As the Theological and mucoadhesive properties of the gels correlate well to each other, the prepared gels were optimised for maximum mucoadhesion force (MDF) using a relationship between the (). The gels were evaluated for physical appearance, pH, in vitro release profiles, microbiological and bioadhesion studies. Mathematical relationship obtained, MDF = 24.87 + 9.87 X_1 + 1.61 X_2 + 0.34 X_3 - 0.35 X_1 X_3 + 0.22 X_2 X_3 - 2.47 X_1~2 + 0.0042 X_2~2 + 0.20 X_3~2 (r~2 = 0.9980), explained the main and quadratic effects, and showed that the X_1 and X_2 are the main controlling factors for the mucoadhesive polyherbal gels.
