This paper presents the preparation method of immobilized enzyme used in thebiosensors to detect pesticide residue based on the principle of enzyme inhibition. Chicken liveresterase enzyme is immobilized by using ion exchange resin as the carrier and glutaraldehydeas the crosslinking agent. The highest immobilized enzyme activity is obtained under thefollowing conditions: the concentration of the enzyme activity is maintained at 6U/ml±2U/ml, thepH value is 6.4, the ion concentration is 0.25M, the adsorbing time is 75min and theconcentration of glutaraldehyde is 0.05%, cross -linking time is 12h at 4°C during the cross-linkingprocess. Furthermore, compared to that of the immobilized enzyme prepared with the adsorptionmethod alone, the stability of the immobilized enzyme prepared with the adsorb-crosslinkingmethod is much better not only during the flow analysis process but also in the preservationstage.
