Mono-PEGylated epidermal growth factor (EGF) was prepared and its biological activity dependent on the sites of PEGylation was also characterized in this study. Tryptic mapping and MALDI TOF analysis found the sites of PEGylation: Lys28, Lys48, and N-terminus, in EGF. Cell proliferation assay and in vitro EGF receptor binding assay demonstrated that PEG-N-terminus EGF conjugate showed the highest biological activity.
