Objective: To investigate the enhancin biological effect of different dosages of microbubble contra! agent on High-Intensity Focused Ultrasound (HIFU) ablatio in goat livers in vivo. Methods: Twenty goats were divided int 4 groups randomly. Animals in group K 2 and 3 were bolu< injected with O.Olml/kg/doK 0.03ml/kg/dot and 0.05ml/kg/dc of SonoVue (Bracco SpA, Milan, Italy) intravenousl respectively before HIFU exposure; and those in group 4 wer not given injections as control. The livers were ablated usin HIFU performed in the manner of a single dot using a clinic: device. The frequency of HIFU was 0.8MHz; the intensity < HIFU was 19000W/cm2.The distance from skin to the targ< liver tissue was 30mm. The exposure time was set at 15S for a animals. All animals were euthanized 7 days after HIFU, an volumes of coagulated necrosis were measured. Pathologies examinations were performed to determine whether ther were residual intact tissues within the exposure region Results: Coagulated volumes in group 1, 2, 3 were larger tha those in group 4, with significant difference (p<0.05). Th coagulated volumes increased gradually from group 1 to grou 3, with significant difference (p<0.05). Pathologic: examinations confirmed that there were no residual unaffecte tissues within the exposed volume. Two remarkable change occurred in one goat in group 3 seven days after HIFU: Th surrounding adjacent tissues outside the reactive zone wei necrotized, and the overlying liver tissues were destroyec Conclusions: These findings showed that the enhancing effet of microbubble contrast agent in HIFU ablation was relate with the dosage of the microbubble contrast agent SonoVui The higher the dosage, the larger the volume destroyed in th target tissue. However, when exceeding a certain dosage leve SonoVue would give a negative impact on HIFU ablatioi which awaits further study in future. Conclusions: Thes findings showed that the enhancing effect of microbubbl contrast agent in HIFU ablation was related with the dosage c the microbubble contrast agent SonoVue. The higher th dosage, the larger the volume destroyed in the target tissui However, when exceeding a certain dosage level, SonoVu would give a negative impact on HIFU ablation, which await further study in future.
