Salivary agglutinin (SAG), lung glycoprotein-340 (gp-340) and Deleted in Malignant Brain Tumours 1 (DMBT1) are three names for identical proteins encoded by the dmbt1 gene. DMBT1/SAG/gp-340 belongs to the scavenger receptor cysteine-rich (SRCR) superfamily of proteins, in the form of either secreted or membrane-bound proteins with SRCR domains that are highly conserved from human down to sponges. DMBT1 plays roles in epithelial and stem cell differentiation as well as innate defence factor acting as a pattern recognition molecule. Our early studies have identified the first SRCR as an HIV-1 inhibitory domain, suggesting its potential as an anti-viral agent. In this study, we reported the expression of N-SRCR, the first SRCR domain and one-half of the first SRCR interspersed domain(SID). The expressed protein bound in a Ca2+-dependent manner to the same HIV-1 V3 sequences previously identified to interact with full-length SAG. The current study laid the foundation for further studies on the SRCR sequence/structures important for HIV-1 inhibition and the design of new HIV-1 entry inhibitors.
