Objectives: To discuss the way to test gene expression in single mouse oocyte and embryos. Methods: The Nest RT-PCR was used to test the Tcll and S16 expression in mouse oocytes and peri-implantation stage embryos. Results: The nest RT-PCR amplification of mouse oocytes, sperms and cumulus cells suggested that both Tcll and S16 expressed in single oocyte, and only S16 expressed in sperm and oocyte. The nest RT-PCR amplification of peri-implantation stage embryos suggested that both Tcll and S16 expressed in pronuclear stage, 2-cell stage, 4-cell stage and 8-cell stage embryos, morulas and blastocyst. Conclusion: The nest RT-PCR can be used to test gene expression in single oocyte and early embryos.
