Development of disulfide cross linked block peptides contain arginine and histidine for siRNA delivery

To develop a multifunctional gene delivery carrier, we prepared the oligoarginine carriers containing histidine, CH_2R_4H_2C conjugated with stearic acid (STR). This carrier could easily make stable complexes with siRNA by positive charge and disulfide cross linkage of cysteine residue. The particle size, z-potential, siRNA condensation ability, cellular uptake ability, in vitro silencing effect, and in vivo anti-tumor effect in S-180 bearing mice were determined.
