In this paper a kind of surface acoustic wave (SAW) biosensor for detecting the gene of Staphylococcal enterotoxin B was established. The Love wave device with the substrate of ST-cut quartz and the guiding layer of SiO_2 was used as the SAW biosensor. Because of its remarkable toxicity and stability, SEB was considered a prime threat as a biological weapon of mass destruction. We investigated a method to measure SEB by detecting SEB DNA in samples with gene biosensor, demonstrating that biosensor-based measurement of SEB DNA was possible not only with simplex SEB DNA samples but also in complex samples in the presence of the other enterotoxins' DNA. Other Staphylococcal enterotoxins' DNA samples did not evoke a positive response. The assay sensitivity was 10ng/ml and the linear ranger was 35~200ng/ml.
