    • 摘要: 目的探讨烟草依赖者静息态功能磁共振(rs-fMRI)下大脑奖赏网络是否会发生变化,并分析吸烟依赖程度与奖赏网络的相关性。方法招募2019年7月—2020年2月于我院保健科和体检中心查体的男性烟草依赖者28例(吸烟组),同时招募与吸烟组相匹配的从未吸烟者27例(对照组)。应用GE3.0 T磁共振对所有受试者行脑部rs-fMRI扫描,对图像进行静息态处理,比较两组受试者大脑双侧奖赏网络功能连接(FC)强度,以及大脑双侧奖赏网络FC强度与尼古丁依赖检测(FTND)分值的相关性。结果两组比较吸烟组奖赏网络FC降低的脑区主要有右侧后扣带回、右侧梭状回、右侧枕中回、右侧小脑前叶和左侧岛脑;增高的脑区主要在左侧额上回、右侧颞叶(P<0.01)。奖赏网络FC强度与FTND分值呈正相关的脑区主要有海马旁回、右岛脑;呈负相关脑区主要在左颞中回和左额下回。结论烟草依赖者的奖赏网络发生了变化,而且这种变化的奖赏网络模式与烟草依赖程度有关。

    • 摘要: 急性和慢性痛时往往伴随着诸如焦虑、抑郁等负性情绪。中脑腹侧被盖区(ventral tegmental area,VTA)、伏核(nucleus accubem,NAc)、岛叶(insular cortex,IC)等脑区均参与对痛刺激的反应和负性情绪的调节。VTA是中脑多巴胺传出投射的起源部位之一,但VTA是否向NAc和IC发出分支投射,尚未见报道。本研究旨在探索VTA是否向NAc和IC发出分支投射及其神经化学解剖特点。先分别将逆标示踪剂荧光金(FG)和荧光微球(FMB)用压力注入5只SD雌性大鼠的右侧IC和右侧NAc;动物存活2周后用4%多聚甲醛溶液灌注固定;再冰冻切片,将切片分为3组收集。

    • 摘要: 目的研究伏核神经细胞的细胞外信号调节激酶(extracellular signal-regulated kinase1/2,ERK1/2在甘丙肽受体1(galanin receptor 1,Gal R1)激活对神经痛大鼠的镇痛作用中的机制。方法大鼠左侧坐骨神经结扎构建神经痛模型。行为学方法测量大鼠对伤害性热/机械刺激诱发的后爪缩爪潜伏期(hind paw withdraw latencies,HWLs);蛋白免疫印迹法检测磷酸化-ERK1/2(phospho-ERK1/2,p-ERK1/2)在大鼠伏核中的表达;并于坐骨神经结扎大鼠伏核内注射Gal R1的激动剂M617和拮抗剂M35后,检测伏核神经元p-ERK的表达。最后在坐骨神经结扎大鼠伏核内注射M617,5 min后伏核内注射ERK1/2抑制剂SCH772984,测定大鼠的HWLs,研究ERK1/2在Gal R1激活对神经痛镇痛作用中的作用机制。结果左侧坐骨神经结扎后第14天(hot-plate test:Pleft0.05);同时坐骨神经结扎后第28天大鼠伏核神经细胞的p-ERK表达上调(P0.05)和第1天差异无统计学意义(P>0.05)。坐骨神经结扎大鼠伏核内注射M617后p-ERK的表达下调(P0.05);最后,坐骨神经结扎大鼠伏核内注射3μg(hot-plate test:Pleft0.05;Randall Selitto test:Pleft0.05)和6μg(hot-plate test:Pleft0.05)。结论在神经痛大鼠的伏核神经细胞,ERK1/2抑制在Gal R1激活引起的镇痛作用中具有重要作用。

    • 摘要: Objective The aim of this study was to investigate the effect of microinjection ibudilast on the appendix pain threshold in nucleus accumbens(NAC)of morphine-addicted rats. Methods Normal rats and morphine-addicted rats were injec-ted with ibudilast(5 μg/2 μL)or saline(2 μL)into NAC through glass microelectrode. The tail-flick period(TFL)was used as an in-dicator to observe the thermal pain threshold before and after ibuilast injection. Results After injected ibudilast 5 μg into the NAC of normal rats and morphine-added rats,the TFL was prolonged compared with that before injection,and the TFL prolongation of mor-phine-addicted rats was more significant than that of normal rats. There was no significant change in TFL after 2 μL of saline was in-jected into the NAC of normal rats and morphine-addicted rats. Conclusion Microinjection of ibudilast into NAC of normal rats and morphine-added rats can cause analgesic effect,and the analgesic effect of morphine-addicted rats is more significant.%目的 研究伏核(Nucleus accumbens NAC)内微量注入异丁司特对吗啡成瘾大鼠甩尾痛阈的影响.方法 正常大鼠及吗啡成瘾大鼠伏核经玻璃微电极注入异丁司特(5 μg/2 μL)或生理盐水(2 μL)后,以甩尾潜伏期(TFL)为指标,观察注药前后的镇痛作用.结果 正常大鼠及吗啡成瘾大鼠伏核内注入5 μg的异丁司特后,与注药前相比,其TFL延长,并且吗啡成瘾大鼠TFL延长较正常大鼠更为显著;Wistar正常大鼠及吗啡成瘾大鼠伏核注入2 μL生理盐水后,TFL无显著变化.结论 大鼠及吗啡成瘾大鼠伏核内微量注入异丁司特后,可使其产生镇痛作用,并且吗啡成瘾大鼠的镇痛效果更为显著.

    • 摘要: Objective To determine the antinociceptive role of gastrodin in nucleus accumbens (NAc)of rats with neuropath‐ic pain .Methods The left sciatic nerve of rat was ligated to mimic neuropathic pain model ,after which the present study observed the effect of intra‐NAc injection of gastrodin on the hindpaw withdrawal latency (HWL)to noxious thermal and mechanical stimula‐tion .Results The HWL in response to thermal and mechanical stimulation significantly decreased in bilateral hindpaws in the 10th day after left sciatic nerve ligation(thermal stimulation:tleft =7 .35 ,P<0 .01;tright = 3 .24 ,P<0 .05;mechanical stimulation:tleft =5 .50 ,P<0 .01 ;tright =3 .32 ,P<0 .05) .Intra‐NAc injection of 40 or 20 μg/μL ,but not 10 μg/μL of gastrodin markedly induced an increase in the HWL in rats with neuropathic pain .Conclusion Gastrodin contributes an important antinociceptive role to neuro‐pathic pain in NAc of rats with mononeuropathy .%目的:构建神经病理性痛动物模型,研究伏核注射天麻素对神经病理性痛的镇痛作用。方法实验采用左侧坐骨神经干部分结扎致神经损伤制作神经痛动物模型,观察伏核注射天麻素对伤害性热刺激和压力刺激诱发的大鼠后爪缩爪潜伏期的影响。结果左侧坐骨神经干部分结扎,引起双侧后爪缩爪潜伏期缩短,尤以10 d后左侧明显(热板测试:t左=7.35,P<0.01;t右=3.24,P<0.05。Randall Selitto测试:t左=5.05,P<0.01;t右=3.32,P<0.05)。神经病理性痛大鼠伏核注射40μg/μL和20μg/μL天麻素引起双侧后爪缩爪潜伏期延长,但注射10μg/μL的天麻素没有显著性影响。结论天麻素在大鼠伏核对神经病理性痛具有镇痛作用。

    • 摘要: 目的 探讨神经病理性痛大鼠伏核注射甘丙肽的镇痛作用及其对星形胶质细胞活化的影响.方法 左侧坐骨神经结扎构建神经病理性痛大鼠模型,伏核注射甘丙肽,测定伤害性热刺激和压力刺激诱发的后爪缩爪潜伏期(HWL),观察甘丙肽的镇痛作用;Western bolt法检测星形胶质细胞的特异性标志物-胶质原纤维酸性蛋白(GFAP)的表达,观察伏核注射甘丙肽对星形胶质细胞活化的影响.结果左侧坐骨神经结扎引起大鼠双侧HWL缩短,且伏核GFAP的表达增加;神经病理性痛大鼠伏核注射甘丙肽引起双侧HWL延长,GFAP的表达减少.结论伏核微量注射甘丙肽对神经病理性痛的镇痛作用可能通过抑制星形胶质细胞的活化来完成.

    • 摘要: 化疗耐药是制约卵巢癌治疗效果的主要原因之一,寻找逆转卵巢癌耐药的有效方法是当前国内外研究的热点。近年来研究表明,伏隔核因子1(nucleus accumbens-1,NAC-1)在卵巢癌中的过度表达不仅在其发生发展、转移复发中起重要作用,而且与化疗耐药密切相关。NAC-1通过调节药物耐药相关的多种信号通路,导致卵巢癌细胞对抗肿瘤药物的敏感性降低。因此以NAC-1为靶点逆转卵巢癌耐药可能为治疗开辟新的思路,NAC-1特异性抑制剂联合其他化疗药物有望成为卵巢癌新的治疗策略。现就NAC-1在卵巢癌化疗耐药中的研究进展进行综述。%Resistance to antineoplastic drugs is a major cause that leads to poor treatment effect in ovarian cancer. Finding effective ways of ovarian cancer chemoresistance reversal has become a hotspot at home and abroad. A rapidly growing number of recent studies shows that, overexpression of nucleus accumbens 1 (NAC-1) in ovarian cancer is not only involved in pathogenesis, progression, metastasis and recurrence, but also closely related with tumor resistance. NAC-1 regulates multiple signal pathways of ovarian cancer chemoresistance to reduce anti-tumor drugs sensitivity. Therefore, the specific targeting of NAC-1 could open new avenues for reversing ovarian cancer chemoresistance, the specific inhibition of NAC-1 activity in ovarian cancer cells combined with chemotherapy is expected to be an effective cancer treatment strategy. This article reviews the mechanism of NAC-1 in ovarian cancer chemoresistance.

    • 摘要: 目的 评价孕晚期吗啡暴露对子代大鼠伏隔核含2B亚基的N-甲基-D-天冬氨酸(NM-DA)受体(NR2B)表达的影响.方法 孕14 d的SD大鼠48只,16~ 20周龄,体重400 ~ 600 g,采用随机数字表法分为2组(n=24):对照组(C组)和吗啡组(M组).M组于皮下注射吗啡,2次/d(9:00 am和5:00 pm),首日剂量为2 mg/kg,逐日递增1 mg/kg,至6 mg/kg后每天不再增加剂量直至孕鼠分娩.C组用生理盐水替代吗啡.每组取体重接近的子代大鼠50只,正常喂养至8周龄.每组取30只子代大鼠,采用恒量法连续皮下注射吗啡(3 mg/kg)7 d进行条件性位置偏爱训练,训练结束24 h时,记录伴药区停留时间.每组取10只子代大鼠,分别采用免疫组化法和Western blot法测定伏隔核NR2B的表达水平.结果 与M组比较,C组子代大鼠伴药区停留时间延长,伏隔核NR2B表达上调(P<0.05).结论 孕晚期吗啡暴露导致子代大鼠吗啡成瘾易感性的机制与上调伏隔核NR2B的表达有关.%Objective To evaluate the effect of morphine exposure in the late pregnancy on the expression of 2B subunit-containing N-methyl-D-aspartate receptor (NR2B) in the nucleus accumbens of offspring rats.Methods Forty-eight Sprague-Dawley rats at 14 days of gestation,aged 16-20 weeks,weighing 400-600 g,were equally and randomly divided into control group (group C) and morphine group (group M).In group M,morphine was administered subcutaneously twice a day (9:00 a.m.and 5:00 p.m.) in the initial dose of 2 mg/kg followed by daily increment of 1 mg/kg until 6 mg/kg.The equal volume of normal saline was given instead of morphine in group C.Fifty offspring rats with similar body weight were selected from the two groups,and conventionally fed for 8 weeks.Thirty offspring rats were further selected,morphine 3 mg/kg was injected subcutaneously for 7 consecutive days,and then the rats underwent the conditioned place preference test to monitor the effect.The time spent in drug-paired side (gray area) was recorded at 24 h after the test.Ten offspring rats selected were sacrificed after the test,and the brain was removed.The nucleus accumbens was isolated for detection of the expression of NR2B by immunohistochemistry and Western blot.Results Compared with group M,the time spent in drug-paired side (gray area) was significantly prolonged,and the expression of NR2B in the nucleus accumbens was up-regulated in group C (P<0.05).Conclusion The mechanism by which morphine exposure in the late pregnancy causes susceptibility to morphine addiction is associated with up-regulation of NR2B expression in the nucleus accumbens of offspring rats.
