The present invention relates to the technical field of biologic formulations, and provides a preparation method for and an application of an exosome derived from human umbilical cord mesenchymal stem cells. The preparation method comprises the following steps: culturing human umbilical cord mesenchymal stem cells to obtain an exosome-containing supernatant; and separating the exosome-containing supernatant by a chemical precipitation method to obtain a protein kinase-containing MSC exosome. The exosome derived from human umbilical cord mesenchymal stem cells prepared by the preparation method of the present invention contains a protein kinase. The protein kinase-containing exosome can enhance the AMPK activity and/or inhibit liver cell steatosis, and thus can treat nonalcoholic fatty liver diseases.
