The present invention relates to a method for simultaneously detecting vitamin K1 and vitamin K2 in a sample, and more specifically, (a) adding NaCl, an extraction solvent and a dispersion solvent to a sample, and dispersive liquid-liquid micro-extraction (Dispersive liquid-liquid) performing microextraction, DLLME) to obtain an extract; and (b) analyzing vitamin K1 and vitamin K2 in the extract by liquid chromatography-atmospheric pressure chemical ionization mass spectrometry (LC-APCI-MS). The simultaneous detection method of the present invention is a factor affecting the extraction efficiency of DLLME, and as a result of optimizing the extraction conditions in consideration of the type and volume of the extraction solvent and dispersion solvent, the concentration of NaCl, etc., the recovery rate of vitamin K1 and vitamin K2 is high. Also, it is characterized in that it is eco-friendly by reducing the extraction time and solvent volume by about 90% or more compared to the previously proposed method.
