The present disclosure provides methods and compositions for enhancing an immune response against cancer and pathogens. This relates to cells containing antigen recognition receptors (eg, chimeric antigen receptors (CARs) or T cell receptors (TCRs)) and dominant negative Fas polypeptides. In certain embodiments, the cells are antigen-oriented, demonstrating enhanced cell persistence and the effectiveness of enhanced antitarget treatment. In certain embodiments, the dominant negative Fas polypeptide comprises at least one modification in the cytoplasmic death domain.
