Process recording slides for immunohistochemical staining

Apparatus and methods are disclosed for determining the efficacy of paraffin removal, antigen retrieval, and primary and secondary staining reagents used in analytical processes, in particular, multi-step immunohistochemistry (IHC) assays. In particular, the device comprises an adhesive coated microscope slide containing a plurality of compounds applied as dots in a 2D or 3D structure and sealed under a paraffin coating. Tissue sections or loose cells are then applied to the same slide, all of which undergo IHC processing steps from tissue collection through application of coverslips. Compounds react with either primary or secondary IHC staining reagents to document the experience of treatment of coexisting tissue sections or loose cells. Primary targets consist of bound antibodies applied in a gradient density array. Two arrays of secondary targets are used, one array of mice and one array of rabbits. Each secondary array is applied to the substrate as a 2D gradient density array of 10% to 100% concentration with a 2D/3D 100% concentration target. Develop a scale (or ruler) to objectively measure the concentration of each antigen on coexisting tissue sections, when assisted by extrapolation of secondary arrays and, if available, primary arrays. IHC staining (staining) experience is permanently associated with coexisting tissue sections or loose cells, supporting objective measurements of the QC and antigen density of the IHC process.
