Disclosed are apparatus and methods for determining the efficacy of paraffin removal, antigen recovery, and primary and secondary coloring reagents used in an analytical process, particularly multi-step immunohistochemistry (IHC) analysis. In particular, the device comprises an adhesive coated microscope slide containing a plurality of compounds that are applied as dots in a 2D or 3D structure and sealed under paraffin coating. Tissue sections or loose cells are then applied to the same slide, all going through an IHC treatment step from tissue capture to application of a cover slip. Compounds record the experience of treating coexisting tissue sections or loose cells by reacting with either primary or secondary IHC staining reagents. Primary targets consist of bound antibodies applied in a gradient density array. Two arrays of secondary targets are used, one array is a mouse and the other array is a rabbit. Each secondary array is applied to the substrate as a 2D gradient density array of 10% to 100% concentration with a 2D / 3D 100% concentration target. A measure (or ruler) to objectively measure each antigen concentration on coexisting tissue sections when extrapolated from the secondary array and, if available, is supported by the primary array. IHC staining (pigmentation) experience is permanently linked by coexisting tissue sections or loose cells to support objective measurement of QC and antigen density in the IHC process.
