The present invention relates to a polymer-based material having a covalently bound and enzymatically degradable peptide sequence that cannot be degraded by the biological and metabolic activities of cells and tissues, each peptide sequence of 2 to 15 amino acids. and incorporated into or bonded onto the polymer-based material. Thus, the peptide sequence may be part of a two-dimensional or three-dimensional structure of a polymer-based material. By enzyme-addition-controlled degradation of covalent bonds within the peptide sequence, bio-orthogonal degradation of the three-dimensional structure is achieved, or at least a portion of the molecule when the peptide sequence is conjugated onto a polymer-based material. release is achieved. The present invention further provides for in vitro production of cell cultures or tissues or organs; for in-vivo stabilization of donor cells, tissues or organs; and to the use of such polymer-based materials for in vivo or in vitro treatment of living cells, tissues and organs. Unless the adhesive bond between the material on the one hand and the sample on the other hand, i.e. a cell, tissue or organ, does not destroy the integrity of the sample (extracellular matrix and cell-cell and extracellular matrix-cell junction) can be controllably degraded (bio-orthogonality).
