Disclosed is a method for detecting tetracycline in milk on the basis of surface-enhanced Raman technique, the method comprising using a nano-silver substrate with a PDMS chamber structure as a Raman enhancement substrate, and using 4-amino-1 butanol in combination with a SERS substrate, and the enhancement effect by using SERS is better than that by using a PDMS chamber or nano-silver particles alone. The method is used to detect tetracycline in milk, without pretreatment of milk samples, the detection time is within ten minutes, and the detection limit is as low as 0.2 μg/L.
