The present disclosure provides binding polypeptides (eg, antibodies) and effector moiety conjugates (eg, antibodies) comprising site-specifically engineered drug-glycan linkages within native or engineered glycans of the binding polypeptide - drug conjugates or ADCs). The present disclosure also provides nucleic acids encoding the antigen-binding polypeptides, recombinant expression vectors and host cells for producing the antigen-binding polypeptides. Also provided are methods of using the antigen-binding polypeptides disclosed herein to treat a disease.
