METHOD FOR CONSTRUCTING SEQUENCING LIBRARY OF AN RNA SAMPLE TO BE SEQUENCED

Disclosed in the present invention is a method for constructing a sequencing library of an RNA sample to be sequenced. According to the method, poly(A) is first added to the RNA by using a poly(A) polymerase; next, reverse transcription is performed; and then a fixed sequence is added to a 3' end of a cDNA as a primer anchor site by using a terminal transferase; library construction can be simultaneously performed on multiple types of RNAs (comprising mRNA, lncRNA, small RNA, cfRNA, and tRNA), and the constructed libraries simultaneously comprise an mRNA library, a lncRNA library, a small RNA library, a cfRNA library, and a tRNA library; moreover, during the process, no special fragment recovery method is required to recover the small RNA library, so that library construction is only required to be performed once for one RNA sample, and information of multiple types of RNAs can be simultaneously obtained, thereby improving the efficiency and simplicity of RNA library construction. The present invention has important application value.
