Disclosed is a micro-nanofluidic chip, comprising at least one nano-channel array layer (30) and at least one micro-channel array layer (40), wherein the nano-channel array layer (30) and the micro-channel array layer (40) are alternately stacked; the nano-channel array layer (30) comprises a nano-channel (32); the micro-channel array layer (40) comprises an inlet unit and/or an outlet unit; the inlet unit comprises an inlet micro-channel array and an inlet (10); the outlet unit comprises an outlet micro-channel array and an outlet (20); the inlet micro-channel array comprises an inlet micro-channel (42); the outlet micro-channel array comprises an outlet micro-channel (46); and the inlet micro-channel (42) and the outlet micro-channel (46) are in communication by means of the nano-channel (32). The nano-channel can be used for effectively and mechanically extruding biological particles with membrane structures, such as extracellular vesicles, so that the membrane perforation and drug loading of the biological particles such as the extracellular vesicles are realized, drug loading efficiency can be effectively improved, biological activity is maintained, and the flux of drug loading is improved. Further provided are a method for preparing the micro-nanofluidic chip and a method for preparing a drug-loaded biological particle by means of the micro-nanofluidic chip.
