Apparatus and method for performing colorimetric or absorption ELISA detection using a microfluidic ELISA reactor. In one embodiment, an opaque mask is superimposed with the reactor. The mask has one or more windows that are aligned over the detection area of the microfluidic channel. A light beam transiting exclusively through the window and detection area is received by a detector for colorimetric or absorption analysis. In another embodiment, the microfluidic ELISA reactor includes a linearly extending microfluidic channel, a lengthwise portion of which comprises the detection area. An opaque mask having at least one optically transparent window is superimposed with the body. The window directly overlays the detection area to facilitate transit of a parallel light beam from a light source lengthwise through the detection area while the region of the body surrounding the ELISA reactor blocks the light beam.
