To provide methods to promote the differentiation of pluripotent stem cells.SOLUTION: Provided are in vitro methods of stepwise differentiation of pluripotent cells to differentiated population of pancreatic endoderm cells and to population of pancreatic endoderm system cells, the in vitro differentiated population of the pancreatic endoderm cells being obtained from the stepwise differentiation of the pluripotent cells, the cells at each step of differentiation being cultured in medium comprising 5 mM to 20 mM glucose, the methods including processes of culturing cells at each step of differentiation in medium comprising 5 mM to 20 mM glucose.SELECTED DRAWING: None
