Combining penicillinase-sensitive penicillins with fusidic acid for use against penicillinase-producing bacteria

Pharmaceutical antibacterial compositions for oral, topical and parenteral administration comprise fusidic acid (antibiotic ZN-6) or a non-toxic salt thereof and a penicillinase-sensitive penicillin antibiotic or a non-toxic salt thereof. Specified salts of fusidic acid include those of sodium and diethanolamine (preferred), potassium, ammonium, calcium, magnesium, triethylamine, piperidine, morpholine, cyclohexylamine, monoethanolamine, dibenzyl ethylenediamine, benzyl-b -phenylethylamine and procaine. The compositions may be in conventional forms, e.g. ointments, aqueous and oily injections, pills, capsules, tablets, enterically coated tablets and tablets in which the active compounds are separated (particularly when the fusidic acid salt is that of a strong base) e.g. layered and "double" tablets prepared from separate granulations, each comprising one of the above types of antibiotic. Optionally, there may also be present antibacterial nitro-furan derivatives and sulphonamides, other antibiotics which are not absorbed in the intestinal canal and sulphamylbenzoic acid derivatives capable of delaying the excretion of penicillins.
