New products of extraction of a material containing constituents of the membrane of yeast cells, their process of preparation and containing the medicinal product

1502902 Yeast extracts BAYER AG 13 Oct 1975 [5 Nov 1974 7 May 1975 12 Aug 1975] 41822/75 Heading A5B [Also in Divisions C3 and C6] Therapeutic products exhibiting tumourinhibiting activities are obtained by subjecting a material containing or derived from yeast cell walls to extraction with phenol-water and recovering the product from the aqueous phase of the extraction. The material extracted may be (1) complete yeast cells, or a proteolytic (trypsin) or autolytic digest of the cells ("zymosan"); (2) yeast cell walls, or a proteolytic (trypsin) degradation product thereof ("yeast cell wall zymosan") or (3) glucomannon or glucan protein derived from the yeast cell walls. The material may have been derived from yeast of the species Saccharomyces cerevisiae or Candida utilis. Freeze-dried baker's yeast may be used. The material is typically extracted with 10 to 100 parts per part of material of an equal mixture of phenol and water by volume at a temperature of from 40 to 100‹C for 10-30 minutes. The therapeutic product is recovered by dialysis of the aqueous phase followed by lyophilisation of the retentate (sediment and/ or supernatant liquor) obtained by the dialysis. The product may be made up into tablets, &c. in unit dosage form or otherwise. Pharmaceutical compositions may be prepared containing 0.5-95% by weight of the product. Suitable adjuvants (conventional) for such medicaments and pharmaceutical compositions are set forth in detail. Examples 16 and 20 describe the preparation from yeast cell wall of glucomannon proteins I and II.
