PURPOSE:Utilizing the catalytic action of pyruvate kinase in the production of adenosine triphosphate in vivo, cardiac infarction is readily and characteristically detected with high accuracy by relatively simple procedures. CONSTITUTION:The sample of serum is treated with substrates of phohsphoenol pyruvate and adenosine diphosphate in a 7.4pH buffer of tris(hydroxymethyl)aminomethane in the presence of magnesium and/or potassium ions and the pyruvic acid formed is converted to the hydrazone with 2,4-dinitrophenylhydrazine under weak acidic conditions and then, the color of the hydrazone is delveloped in an alkaline medium. The light absorption of the hydrazone is measured (As) at a wave length of 510nm and at the same time the normal value (Abl), blind test value (Ao) and the absorbance (Ast) of the standard solution of 2.0mmol lithium pyruvate are measured and the pyruvate kinase activity value (PKV) is calculated from them using the following equation.
