STAINING AND ANALYSIS OF BACTERIAAbstract of the DisclosureA bacterial staining composition and methods ofanalysis of both gram-negative and gram-positive bacteriaare disclosed. The composition comprises a chelating agentand a basic dye, both of which are operative at a pH aboveabout 7Ø Bacterial staining may be effected by contactingeither concentrated or fluidly suspended bacteria with thecomposition at a neutral or basic pH. Bacteria stained withthe composition and concentrated by filtration, centrifugationor the like are readily visible and their presence in a specimenmay, thus, be rapidly detected. The gradations of color of thestained, concentrated bacteria correspond to the number ofbacteria and semi-quantitative analysis of the bacteria maybe effected by comparison with a standard. Differentiation ofgram-negative and gram-positive bacteria may be accomplished bytreating the stained bacteria with an organic acid wash havinga pH of about 2.5 to 2.6. Such a wash completely decolorizesonly stained gram-positive bacteria. Finally, a method fordetermining bacterial susceptibility to antimicrobial agentsis provided which comprises incubating bacteria with an anti-microbial agent, staining the bacteria and thereafter comparingthe color gradation of the stained, concentrated bacteria witha control or standard.
